I've been decalcifying rat teeth and then frozen sectioning them. I've decalcified for 3 weeks in EDTA with daily changes. My samples do not seem to be cutting very well. They contain a lot of artifacts that are interfering with my analysis. It looks like the dentine is fracturing and the borders of the tooth seem to be tearing. Any ideas on what this might be caused by?
I've been told that I need to decalcify for longer but I am already deacalcifying for a long time. I was also told that it might be a fixation issue (is there such a thing as over-fixation/what does it look like?)
