Sonication Help!

Hi everyone,

I've been searching endlessly for a protocol too sonicate genomic DNA using a cup horn (ultrasonic bath with circulating water to cool) to an average fragment size of ~150-600bp for methylation capture (Invitrogen Methyl Miner).

I'm using the Misonix 4000 (Brochure: http://www.sonicator...00_Brochure.pdf ) The cup we are using is on page 5 (its either 2 or 3).

All my samples were isolated from tissue (liver, pancreas, muscle) and processed with the Qiagen DNA/RNA/Protein Minikit (basically the same as DNAeasy), and have average concentration of 30-90ng/uL (Nanodrop ND-1000) suspended in 100uL of TE buffer.

Any advice or references would be greatly appreciated!! Thanks in advance :)
