It depends on the purpose of your study. If you want to look for expression specifically in islets and you can isolate islets, why not just using islets for RNA isolation?

Yeah, indeed is better to use islets..
But it is kinda troublesome to isolate RNA from islets than pancreas. I am wondering will there be great difference of the gene expression using islets or pancreas?
