My Question would be which enzyme do we need in replicating Plasmids in E.coli? Any idea.

This sounds suspiciously like a homework question... so here's a hint rather than a full answer:

Exactly...Helicase and a DNA pol are at least required.

I don't know what you mean by a big deal, but if you were referring to my reluctance to give a full answer - I do that so as to make you think, it helps you learn and to become a better scientist...
