i have isolated the microvascular endothelial cells from tumor tissue. i wanna cultivate and propagate them. but these cells can only survive for 5-7days, and then they come to senesce gradually. i use several kinds of culture medium supplemented with growth factor. still there is no use. can anyone help me?

Hi ,
I have developed a myoblast culture for cellular cardiomyoplasty. The cells are contaminated with fibroblast from the fascia. All the community use CD56 as a myoblast positive marker but in my flow cytometry's experiments with this antibody, my negative control with dermal fibroblast resulted positive for CD56 too.
Please help me!!
