Can I perform ChIP from FROZEN human buffy coat?
We’re having problems with our assays and we’re thinking that they may be because our starting material is frozen buffycoat (white cell and erythrocytes).
We defrost it, incubate with a red cell lysis solution and precipitate the white cells (sometimes the pellet is still red). Then we start with formaldehyde fixation, shearing of chromatin ,immunoprecipitation etc……
Can we improve our assay. The problem is that we can't change our starting material.
