Hi I am looking for a reliable way to extract histones (without disturbing their methylation state) from brain tissue? I am also getting nuclear fractions to investigate methylation differences by western blot but thinking that extracting the histones themselves from the nucleosome may yield cleaner western results. Any and all opinions on this are very much appreciated, thanks!!

Chromatin immunoprecipitation (ChIP) might be what you are looking for.
