the male and female templates are old whereas other unknown templates were new.
It was stored at 4 degrees because i use it everyday.

To be sure I'd check the DNA with a gel.
And if you replaced all reagents without success, perhaps the thermal cycler is not working properly, or someone changed the program (accidentally hopefully)?

i checked the DNA with the nanodrop though and it was fine, but yes i would check the DNA with the gel also.
As for the thermal cycler program change - i double check it to make sure its the right program.

I checked the DNA with gel, they are fine.
