Hi all
I am new to plant transformation and recently have been working on tobacco leaf disc transformation with LBA4404 strain harboring plasmid with plant selection marker as kanamycin.
I have been following the standard procedure yet I never get any result. Please help me out as i feel frustrated as tobacco being a model system is not being effective.
The selection media i use comprises of MS supplemented with 1mg/l BAP and 0.1 mg/l NAA with kanamycin (100mg/l) and cefatoxime (500mg/l) which is same for shoot bud induction.
Suggestions are welcomed.
