How to justify the validation with southern hybridization over PCR amplification for a specific gene detection ? I know this is a conventional question but still need to look for a better clarification.

Typically Southern blots are more specific as you use a long probe to detect the gene, however, the technique is relatively low sensitivity. PCR has lower specificity but much higher sensitivity.
