Hi everyone!
We are screening some compounds on their immunomodulatory effect. For that purpose we use murine splenocyte cultures and human PBMC cultures in 96 well plates. We have the next stimuli: aCD3+aCD28, PHA, ConA, PMA/Ionomycin and LPS. The cytokines in the supernatants are measured with ELISA, but I'm somewhat confused which cytokines to measure (and which stimuli to use). We don't want to miss any specific cell responses. Is my following suggestion a good one?:
IL-2 specific for T-cells
IFNg specific for B- and T-cells
IL-4 specific for B-and T-cells
IL-6 specific for monocytes and dendritic cells
TNF-a specific for monocytes and dendritic cells
The stimuli:
aCD3+aCD28/PHA/ConA: direct stimulation of T-cells
PMA/Ionomycin: direct stimulation of B- and T-cells
LPS: direct stimulation of monocytes, dendritic cells and B-cells
