Hi everyone,

I have to clone a ~12 Kb fragment into an E. coli plasmid.

So far I have used a p15a ori - based vector, but the plasmid seems to be not very stable.

Does anyone knows whether p15a ori can replicate a 15Kb plasmid?
Is there any better alternative (eg a BAC?).
