We are trying to injection Venus RNA into zebrafish embryos and so far we have had no luck. I am certain we are making RNA correctly and we have been trying to inject at 50picomol total concentration. Has anyone run into any issues injecting Venus and having expression? Are we injecting at too low a concentration? We have used the kit that we make the Venus RNA from with other constructs and they work just fine, so it seems unlikely to be the kit. We have also tried several different Venus constructs with no luck. We have ruled out the injection buffer, water and everything else in common. Any suggestions would be great. Thanks!
