Hi

Does anyone have experience with transfection (electroporation) of the murine macrophage cell line J774.2?

I am planning to use the Neon Transfection System (Invitrogen), and I would be interested in knowing what electroporation parameters to use. I know I can always run an optimization protocol and identify optimal parameters, but maybe someone out there has some experience on this matter already..?

I would have a look on the invitrogen website for the neon system - there are a lot of cell lines' conditions listed on there (but not J774.2, though they do have "macrophage listed).  You may be able to come up with a consensus on the conditions that will be fairly close to optimal.
