hello all
I have a question regarding competent cell formation of E.coli TOF 10
1.Can we use ddH2O to dissolve the pellet to form competent cell rather than Using Cacl2. Since I am and in my lab I saw people using ddH2O instead of Cacl2 but I have never found a protocol using water,
Kindly tell me about this..

Follow the protocol you have for top10, these things have all be worked out for you!  Note that the protocol you are actually using is just a general one for competent cells rather than TOP10 specifically.

Different protocols for different preparations... water is often used for electrocompetent cells.
