When I did the transformation of LR DNA into DH10Bac cells, I used a destination vector (pDest8) as a negative control. However, I saw both blue and white colonies on the plate. Then I pick a white a colony did the plaque purification on the plate containing same antibiotics, incubating for 24 hours at 37 celsius. I saw there are lots of white colonies!
I cannot figure about an reasonable explanation about that... Is there anyone could help me??
