Hi,

I have to do an assay where I need quench metabolic activity of cells infected with a parasite, lyse the cells and isolate the parasite from the host cells to study a particular  parasite metabolite.

Generally I quench metabolic activity using chilled methenol : water (80:20). However the solvent is not sufficient to lyse the host cells instantaneously. what detergent (Or any other compound) should i add to the chilled solvent to lyse the host cell and quench the metabolic activity simultaneously.?????

I have tried DMSO, Triton X100 and Tween. Nothing works at the temperature i want to maintain. :(
