Hello everybody,
I have been doing 2D gel electrophoresis for my yeast protein to identify its posttranslational modifications with a combination of 7M urea and 2M thiourea in my IPG rehydration buffer (at room temperature), unless I came through carbamylation as an artifact from urea. I am afraid if my protein undergoes carbamylation in vitro. I read that guanidine hydrochloride and/or phosphates can be used to avoid carbamylation in vitro. Does anybody knows and can suggest the composition of buffers containing guanidine hydrochloride and/or phosphates?
