Dear friends!
I need your advice regarding the engineering of di-sulfide bond.
The task is following: I have a complex of 2 proteins. I would like to increase it's stability bu introducing an artificial di-sulfide bond between two proteins. I know approximately the residues of both proteins that should be located in proximity to each other. I know how to make site-directed mutagenesis. Could you recommentd any reading on this topic? I.e. protocols, research articles etc etc. or share your experience? What should be the expresssion conditions in this case?
