I cultured cells in a small transparent polymer well. but have to cover the well with parafilm to prevent contamination and evaporation. When I use 10X and 20X objectives on an inverted nikon microscope to take pictures, the contrast of the cells is really weak. Is there any suggestions on how to enhance the phase contrast of the image? Thanks a lot for helping.
