Hi,

I am new to Gateway cloning. I cloned my gene on entry vector via regular subcloning. I did LR reaction with my entry vector and destination vector. When I sequenced my destination vector, I had the right sequence but out of frame! however, the insert on the entry vector seems to be in frame!!!

Any ideas??? is there any considerations I need to know? probably regarding designing primers to do subcloning?
