I tried transforming BL21 DE3 comp cells with pGEX4t2 vector for a control experiment, but after plating the transformed cells on ampicillin LBplates, more than 100 colonies were observed but on picking them and growing them in LB broth with100ug/ml ampicillin growth was observed but no plasmid was detected on running a gel for checking the plasmid.
