Hi,
I encountered problem during the selection for my transformant arabidopsis plant in Kan selection plates.

I grew my wild type and transformation seeds on 50ug/mL Kan selection plates.
I put them on 4 degree C for 2 days, light for 1 day, dark for 2 days and light for the rest.
I found that all my seedlings turned green even for the wild type which suppose to be pale yellow.
Anyone having idea or similar cases for this? How am I going to select this seedlings: based on the bigger size of leaves?? or just leave the seedlings in the plates until the non-transformant one die off?? or should I increase the antibiotic concentration???
