Hi all, I have been trying to standardize sonication of formaldehyde crosslinked (for 15 min) discs and cns collected from drosophila 3rd instar larvae. I am using Bioruptor for sonication (20 min 30 sec on/off), but I do see variations either 100-500 bp or 600-1500 bp. I changed all possible factors (e.g. TPX tubes, crosslinking time, changing cell number etc.) but of no use. Please suggest me, I want 200-500 bp fragments as I want to do Illumina sequencing.
