Dear Guys,
I have RPMI 8226 (Myeloma cell line) I bought from Cell bank
I use RPMI 1640 with 10% FBS
Upon thawing viability was 85%
I have only one incubator with 10 % humidity, in protocol no words about if I should use humidity or not only the Co2 level control only.
I put my medium in incubator for a 15 mins and it was slightly pink.
I added my cells
Next day???
I see the cells rounded , not attached to the surface, but I feel like it is adherent to surface.
Like it is glowed to the surface of flasks (circular not Crescent ).
I tried to determine the viability after 24 hrs culturing and it was 25%, decreased.
Guys???
any advice??? shall I use incubator with no Humidity is that the reasons???
Is it normal as I have never use suspension cells before, and I am almost new in cell culture (5 months)
