hi,

I have exprienced some problem in expressing a human IgG in animal cells.(HEK293 or CHO-K1)

I have checked my gene sequence in the plasmid after ligation by sequencing.

It is correct n in correct reading frame.

IgG is 150 kDa, but expression check by SDS-PAGE and western blot band size was about 100 kDa.

other IgG was expression well same vector system and same cell line. but, this IgG does not 150 kDa complex.

I guess that protein was heavy chain dimer or other complex.

I would really appreciate it if anyone has any suggestions to solve this
