Hello,
I do in-vivo gain-of-function studies with Drosophila microRNAs. I have mutated the seed sequence of a particular microRNA gene and have made transgenic flies that express that mutated transcript. Of course, the transcript doesn't exactly make the same stem-loop structure as its parent. Therefore, the Dicer-1 cutting site might be different or none!
Can RT-PCR (with specific primers) tell me if that putative mutated microRNA exist?
Any input is highly appreciated!
