I am trying to express a chimeric fusion protein in mammalian cells (293T, DF-1, BSC-40) using pCDNA 3.1 using Lipofectamine 2000. However there seems to be no expression of my protein. I used the same sequence to purify protein from E.coli and generate recombinant virus (infecting mammalian cells), and both worked with high level of expression. Could anyone suggest a good solution? Thank you.
