Requesting basic protocols on cell sorting

Hi everybody,

I have to separate epithelial cells in urine by FACS. I have a good labelled antibody for a cell surface marker but I don´t know anything about cytometry and cell sorting. That is why I would be very grateful if someone could advise me on how I should label the cells with this antibody. I normally use PBS-tween for Northern-blots or ELISAs but in this case, I want to recover the cells intact. So I guess I have to avoid detergents, right?

Every FACS machine has a protocol for that. Your machine must have a manual. Have you studied yours?
