Hi,
I am new in miRNA research and have little molecular biology experience. I want to test a miRNA knowdown effect on its targeting gene by RT-PCR.
However, I don't know the standard for designing primers for such usage. My doubts are as follows:
1) should the primers be up and down stream of the cleavage site of the miRNA on the gene? How far should that be?
2) what is the length of the PCR product? How can I get such information?
My questions may sound really naive. Thanks a lot!
