Has anyone had any success with using a FACSCalibur to isolate and grow primary cells (i.e. primary muscle cells)? Some previous members of our lab have tried sorting with the FACSCalibur before but found the sample was too dilute, even after a large amount of centrifugation.
We don't have the cell concentrator option but when I looked it up in "Practical Flow Cytometry, 4th Edition" they seem to allude that recovery of viable cells from it is unlikely. We do have a Millipore stirred cell concentrator though. Has anyone used this to concentrate a cell suspension?
Thanks in advance!
