Hi, I want to compare the 16sRNA level of a certain bacteria between patients and healthy by Real Time-PCR. I am a new starter and hope to get some advice.

1. How can I find the 16srRNA to design pathogen specific primers?

2.what general procedures should I go through?

3. My interested bacteria is a G+. How should I extract the bacteria DNA? The sample contains mammal cells, G+ and G- bacteria.
