Dears,

I typically  store the supernatant of transformed baterial overnight cultures at minus 80 with 10 % DMSO.Hence, it would be grateful to know can i directly take some fraction of it  for an overnight culture and midiprep or better to perform a streak plate and select a single colony, starter culture , followed by overnight culture.

I'd recommend streaking on a plate and growing from a single colony.
