Prolem in screening of hygromycin concentration

hi there,

I have a problem in screening for hyromycin concentration on Ganoderma sp. I have cultured the Ganoderma sp. on PDA with different concentration of hygromycin B. Starting with control, 10ug/ml up to 50ug/ml. The cultures are left in dark at 30C until growth is observed. However, no growth in either plates are observed. What would have gone wrong. Since my control plate is not growing also. Pls help!

It could be anything - but it almost certainly isn't to do with the hygromycin. I would check the medium formulation, the viability of the spores, and the incubator.
