Hi

I wanted to know how do you design antisense oligonucleotide (ASO) for a particular gene? This is in terms of rat transfection. I know how to design primers for cloning, and I thought designing ASOs would be similar to thatPosted Image ...but I am facing problemsPosted Image . I thought the easy way to learn would be from any of the paper, just take the sequence from NCBI and compare the ASO sequence with this. SO for this I followed this paper-

paper is attached also. I found the sequence of p65 unit (RELA) on NCBI but I am not able to figure out how these primers have been designed Posted Image Posted Image ....please help me. Also how do we get the exon-intron sequence on NCBI? That could also be one reason, coz I am not sure whether I have matched the primers with correct coding sequence.

please please help me.....
