hi!

lately, i found(predicted?) nov. gene in plasmid from Soil bacteria(sphingomonas sp.)

first of all, i want isolation that gene, but i was not working in e.coli(still trying!).

and i had to single point mutation of that gene

but, due to the plasmid size(it is 500kb!!), i can not control that anymore.

So, i want get some information about homologous recombination between plasmid and plasmid to control that gene.

if you have any experience, please let it know

or, anyone have some idea of contol that gene
