hi, I am going to use the Xcm I to cloning my gene. but this is enzyme will produce DNA fragments that have a single-base 3' extension which are more difficult to ligate than blunt-ended fragments. and it is the only enzyme I can use. so do you think it will be difficult to clone in? I appreciate any suggestion.
