I am having lots of problems with my IHC staining of human skin for CPDs(cyclobutane pyrimidine dimers). These are paraffin slides. I am using NaOH and Tris HCl for denaturation. Using DAB for staining. My problem is my nuclei are not taking the stain rest everything in the epidermis is all stained heavily. These are sun exposed samples where I should be having tonns of positive nuclei.I have tried different concentrations of the antibody, different incubation times, almost everything. The same protocol had been working previously!!! any help is most welcome!!!
