I co-transfect my cells with my shRNA and the target plasmids by the ration of 0.25:1, 0.5:1, 1:1 and 2:1. and i find the lower shRNA: target ration the better inhibit efficiency. How to explain this? Thank you.

Probably too much shRNA expression has a negative impact on small RNA function by saturating the small RNA processing pathway and Ago proteins.
