Dear All,
I have a plasmid with a short hairpin-RNA sequence in a pRS vector. I want to amplify this vector in bacteria but it is not amplifying whatsoever. It is ampicillin resistance. I am using GC10 E.Coli from Sigma and LB broth. I have read here about using low temperature growth cycle and chloramphenicol. Can anyone suggest the best way to improve yield of this plasmid?
