Hi
I want to ask you what is the best anti-phospho ERK1/2 antibody for western blot. I ussually used one from Sigma (cataloge number: 9692), but it is just doesn´t working anymore.
Like I heed to all the steps of my protocol, I think i have a problem with the antibody, and that is why I would like to know what is the best antibody for detect activated diphosphorylated ERK1/2
PD: i work with mouse T cells lysates.
Thanks!

The rabbit monoclonals from Cell Signaling are excellent. Try one of these:
http://www.cellsigna...ducts/4370.html
http://www.cellsigna...ducts/4377.html
http://www.cellsigna...ducts/4376.html
They all indicate they detect dually phosphorylated ERK1/2 and also singly phosphorylated ERK1/2 (either the Thr or the Tyr depending on the antibody). This was likely the case with your Sigma antibody as well.

Try this one http://www.scbt.com/...4-antibody.html
Way cheaper than Cell Signaling and it works great!
