I am going to isolate and culture primary mouse brain microvascular endothelial cells. I am confused why someone culture them in DMEM with high glucose concentration, the others dont use glucose at all.
In addition, could anyone tell what difference between methods of isolation endothelial cells vs astrocytes?
I am new at tissue culture and any advice for this situation would be well appreciated.

i actually don't know much about primary culture, but it's important to remember that physiological concentration of glucose is 5 mM. cells tend to do better in high glucose, but it mimics hyperglycemia! i don't know if that helps
