Hi, I am wondering what I should use to dilute the lysate that came with my Luciferase kit. Do I use TE buffer? Or DI water?

You could use something from the or check the kit manual. I have used PBS, but do you need to dilute the samples?

I would strongly recommend that you dilute the lysate with additional lysis buffer. This will keep the amount of the diluent components the same regardless of dilution factor of the sample. Addition of other buffers or components could resutl in the addition of a variable amount of an inhibitor or other factor that impacts the luciferase reaction.
