Here is a  robust multi-gene cloning method developped in our lab. it is easy to use and require nothing special. The efficiency is much higher than that of restriction-ligation based methods. http://dx.plos.org/1...al.pone.0030267

This is very nice work.  I would be interested to know your approach to repetitive sequence assembly, such as a construct with two closely related variants of GFP, for example.

Really interesting. I would like to see how they behave with viral vectors construction where there are inverted repeats like in AAV or lentivirus plasmids.
