I am performing a southern blot on gDNA but have failed to get a signal with my probe. i am beginning to think it may be the probe (900bp). i was wondering what housekeeping gene probe i can use just to make sure the problem technically the southern is working. Will gapdh work? thank you

Hi,
if you just want to know if your blot technique is working i'd recommend dot blots with your labelled and unlabelled probe and a probe you know for sure that it's working (i used my commercially available DIG-labelled lambda-marker). Worked well for me during colony filter lifts to find out that my dNTP-labelling mix did not work.

Henry
