I am at the beginning stages of transfection and have added my transfection reagent and dna to my cells.  My question is, now that I want to do my selection with g418, how often do I have to change my media?  My media that i use contains 250ug/ml of g418.  Once a week?  Every other day?

generally when i perform a transfection (g418 selection) i plate the cells with media which doesnt contain g418. This is so the cells can propagate. generally i will do this for 24-48 hours -depends on how the cells look-. i then add media with the selection drug -g418- and immediately i begin to see cell death. i change media every other day until clone screening is complete.
