I would be happy if someone can advise me regarding this issue. So I inserted my cdna of interest into pCDNA 3.1 B (+) his-myc vector to perform some expression studies. I included the Kozak sequence and sequenced to make sure its in frame. The problem I am having now is, I am performing WB to make sure my protein is expressed using a myc antibody I bought from SC. Unfortunately, I am getting a same size band both for my construct and the empty vector. Is anyone encountering similar problem? Any suggestions are welcome.
