Hello, yesterday i was going through papers for calculating Ta (annealing Temperature) of my Allele specific primers.The equation is

Ta=.3xTm(primer) + .7xTm(product) - 14.9

here Tm(primer) is the Tm of the least stable primer-template pair. I want to know what does primer-template pair actually mean and how it should be calculated.
