We are interested in comparing the impact hypoxia and glucose deprivation has on calcium release from the ER. We are going to use the mag fura 2 probe. The issue is we want to measure calcium in real time, meaning in hypoxia because we are concerned that reoxygenation will change calcium levels. My question is is there a way to quench the reaction before readiong the sample, and is there a protocol that is recommended for lysing before reading in the fluromax plate reader.
