My experiment design is;

1. Activate Jurkat T cells with 50 ng/ml PMA for 20h

2. replace media

3. infect cells with virus

4. replace media after 1h adherence time

5. incubate for 2d

6. protein prep


Qs.

I wonder how long the Jurkat cells are activated for upon PMA?

Whether it will last for 2 days after first media replacement (step 2)?

Would I need to re-inject PMA after each media replacement to maintain the effects of PMA (i.e. maintain T cell activation) ?

Also, would this be toxic to the cell??
